Mr. Brooks tells the story of Earl Brooks, a highly-respected businessman who is also a serial killer known as the Thumbprint Killer. Despite his successful facade and recent recognition as Portland's Man of the Year, Brooks struggles to keep his dark secret hidden.
After attending AA meetings and managing to keep his addiction to killing under control for two years, Brooks's alter-ego Marshall re-appears and urges him to kill again. When Brooks gives in to his urges and murders a couple caught in a vulnerable moment, he is photographed by someone with a death and murder fetish.
Meanwhile, a police detective investigating the murder is facing her own challenges. She is going through a messy divorce and is being pursued by a violent criminal who had previously threatened her and has now escaped from prison.
